Acellular buffalo small intestinal submucosa and fish swim bladder for repair of full-thickness. skin wounds In rabbit Vineet Kumar, Naveen Kuma..-, A. K. Sharma, S. K. Maiti, A.K.Gangwar Himani Singh, Sameer Shrivastava*, Sonal,* and R. B. Rai** Division of Surgery *Division of Veterinary Biotechnology, **Division of Pathology Indian Veterinary Research Institute lzatnagar-243122i Uttarpradesh, INDIA 'corresponding author E r:nail: [email protected] ABSTRACT In the present study acellular matrix from small intestine of water buffalo (Bubalus bubalis) and swim bladder of fresh water fish (Labeo rohita) were prepared. using ionic biological detergent. The small intestine treated with 2% ionic biological detergent for36h under constant agitation showed complete loss of cellularity. The mucosa was completely acellular, .submucosa had normal a~rangement of collagen fibres, and muscular layer was acellular with normal muscle cfibre structure. Histological examination confirmed the acellularity of the matrices. The nucleus' and· cytoplasmic cellular components were successfully removed with no evidence of remaining cells, nuclei or other cell·fragments and resulted into acellular intestinal matrix consisting of primarily collagen with good tensile strength. The swim bladder treated with 0.5% ionic biological detergent for 24h under constant agitation showed complete loss of cellularity. The tunica externa and interna were completely acellular. Contrary to native ·tissue the collagen fibres were loosely arranged. Acellular swim bladder was cross-linked with 1,4-butanediol diglycidyl ether (BDDGE) for 48 h and acellular intestinal matrix was cross-linked with 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride (EDC) for 48 h. The prepared acellular buffalo small intestinal submucosa and fish swim bladder were evaluated in the repair of full-thickness skin wounds in rabbits. Thirty (30) clinically healthy adult New Zealand White rabbits of either sex were randomly divided into five groups of 6 animals each to test the efficacy of uncross-linked and cross-linked acellular intestinal matrix and fish swim bladder graft for the repair of full-thickness skin defects. In group A, acellular intestinal matrix, in group B cross-linked acellular intestinal matrix, in group C acellular swim bladder, in group D cross-linked acellular swim bladder and in autograft group (E) self skin was used to repair 2x2 cm 2 size full-thickness skin defects created on both paramedian sides on dorsal aspect under general anaesthesia. The postoperative period was uneventful and there was no evidence of any clinical complications related to the implanted materials. The wounds were examined clinically, immunologically and histo-pathologically at post-implantation days 3, 7, 14, 21, 28 and 42. Clinically, all the animals of different groups were dull and depressed for the first 2 post-operative days. The rectal temperature significantly (P
