Effect of VU0463271 on GDP frequency and amplitude in perinatal mice and rats (related to Figure 1) Quantification of the effect of VU0463271 (10 µM) on normalized frequency and amplitude of GDPs. In P0-2 rats and KCC2+/+ mice, VU0463271 caused a marked increase in both GDP frequency and amplitude (P0-2 rats: frequency fold change 2.30 [IQR 1.39-3.23], p = 0.001; amplitude fold change 1.16 [IQR 1.06-1.30], p = 0.005. KCC2+/+ mice: frequency fold change 1.86 [IQR 1.38-2.43] p = 0.003; amplitude fold change 1.33 [IQR 0.99-1.80] p = 0.033). In P5 rats only the change in frequency was statistically significant (frequency fold change 1.91 [IQR 1.79-2.56] p = 0.018; amplitude fold change 1.15 [IQR 1.02-1.70] p = 0.063). In KCC2-/- mice, neither frequency nor amplitude of GDPs were affected by VU0463271 (frequency fold change 1.00 [IQR 0.66-1.16] p = 0.176; amplitude fold change 1.06 [IQR 0.88-1.18] p = 0.508).
