1. Supplementary Figure 1. Ectopic expression of FOXM1 suppresses the ... scatter (cell complexity; SSC; Y axis) and K1 expression (X axis) 3 days after.
Supplementary Figure 1. Ectopic expression of FOXM1 suppresses the differentiation response caused by loss of p53. a: Immunodetection of FOXM1 and p53 in keratinocytes infected with shp53-1 (shp53) or the corresponding empty vector (plK01, CT1) and FOXM1 (FOX) or corresponding empty vector (plVX, CT2) as indicated. Scale bar: 50 μm. It completes Figure 1a. b: Representative flow-cytometry histograms for keratin K1 in keratinocytes 3 days after infections as indicated. Negative isotype antibody control is represented by red broken line. c: Dot plots showing side scatter (cell complexity; SSC; Y axis) and K1 expression (X axis) 3 days after infections. Positive K1 cells are within red boxes, according to isotype control: B=Basal cells initiating differentiation; S=Suprabasal differentiating cells. Note that the decrease in K1 in shp53/FOX keratinocytes is mainly in the onset of initiation of differentiation (b and c complete Figure 2b).

Supplementary Figure 2. Ectopic expression of FOXM1 alleviates the mitotic block caused by loss of p53. a: Cell cycle of keratinocytes 3 days after infections with CT1 or shp53-1, and CT2 or FOX, by flow-cytometry of propidium iodide (PI) staining for DNA content. It corresponds to quantitations in Figure 3b. b: Percent of cells in the G1 and S (2N), G2/M (4N) or polyploidy (> 4N) phases of the cell cycle, 3 days after infections as indicated. It completes Figure 3b. c: Percent of Brdu positive cells in the different phases of the cell cycle as in b (double staining with PI), 3 days after infection with CT1 and CT2 or CT1 and FOX. It completes Figure 3c. p-values: *
